Product Introduction

Interleukin-2 (IL-2) is a 15-17 kDa pleiotropic cytokine primarily produced by activated CD4+ T cells, CD8+ T cells, and dendritic cells. IL-2 plays a central role in the adaptive immune response by promoting the proliferation, differentiation, and survival of T cells, B cells, and natural killer (NK) cells. IL-2 signals through the IL-2 receptor complex consisting of the IL-2R¦Á (CD25), IL-2R¦Â (CD122), and common gamma chain (¦Ãc/CD132) subunits. At low concentrations, IL-2 preferentially expands regulatory T cells (Tregs) due to their constitutive high expression of the high-affinity trimeric IL-2 receptor, whereas at higher concentrations, IL-2 drives the expansion and effector function of CD8+ T cells and NK cells. Recombinant Mouse IL-2 is extensively utilized in immunology research for T cell expansion and culture, in vitro Treg generation and maintenance, NK cell activation, and preclinical studies investigating IL-2-based immunotherapies and autoimmune disease mechanisms.

Storage and Stability

Protocol (For Reference Only)

Important: Centrifuge the vial briefly before opening to collect lyophilized contents. Reconstitute under aseptic conditions. Avoid vigorous vortexing.

1. Reconstitution: Briefly centrifuge the vial to collect the lyophilized powder. Reconstitute Recombinant Mouse IL-2 in sterile deionized water or sterile PBS to a concentration of 0.1-1.0 mg/mL. Allow the solution to sit at room temperature for 10-15 minutes with gentle swirling to ensure complete dissolution.

2. Dilution for Working Solutions: Following reconstitution, dilute the stock solution in cell culture medium or buffer containing carrier protein (e.g., 0.1% BSA) to prevent non-specific adsorption to plastic surfaces. Do not dilute below 1 µg/mL without carrier protein.

3. T Cell Proliferation and Expansion: Supplement mouse T cell culture medium with Recombinant Mouse IL-2 at a final concentration of 1-20 ng/mL for CTLL-2 cell proliferation or 5-50 ng/mL for primary mouse T cell expansion and maintenance.

4. NK Cell Activation: For NK cell stimulation, add Recombinant Mouse IL-2 to culture medium at a final concentration of 10-100 ng/mL, alone or in combination with IL-12 or IL-15, and incubate for 24-72 hours before assessing cytotoxicity or cytokine production.

5. Treg Expansion: For mouse Treg generation or expansion, supplement cultures with Recombinant Mouse IL-2 at a final concentration of 2-10 ng/mL in combination with TGF-¦Â and anti-CD3/CD28 stimulation for in vitro Treg differentiation, or at 100-500 IU/mL for ex vivo Treg expansion.

Precautions

1. Reconstituted protein is less stable than the lyophilized form; aliquot and store at -80¡æimmediately after reconstitution.

2. Avoid repeated freeze-thaw cycles as this will significantly reduce biological activity.

3. For dilutions below 1 µg/mL, always add a carrier protein (0.1% BSA or 10% FBS) to prevent non-specific adsorption of IL-2 to container surfaces.

4. Optimal working concentrations should be determined empirically for each cell type and experimental system.
