Recombinant Human Interleukin-1 beta, CYT-P01584
Product Introduction
Interleukin-1 beta (IL-1¦Â) is a non-secreted proinflammatory cytokine produced mainly by activated macrophages, as well as neutrophils, epithelial cells, and endothelial cells. It possesses metabolic, physiological, and hematopoietic activities, and plays a central role in the regulation of immune responses. Both IL-1¦Á and IL-1¦Â bind to the same receptor and have similar but not identical biological properties. Mature human IL-1¦Â shares 96% amino acid sequence identity with rhesus and 67%-78% with canine, mouse and rat IL-1¦Â.

Product Features
1. High Purity: >97% by SDS-PAGE and HPLC analyses.
2. High Biological Activity: ED50 <1.0 pg/mL as determined by a cell proliferation assay using murine D10S cells, corresponding to a specific activity of >1.0 ¡Á 10⁹ IU/mg.
3. E. coli Expression: Produced in Escherichia coli as a single non-glycosylated polypeptide chain containing 153 amino acids.
4. Low Endotoxin: <1 EU/µg as determined by LAL method.
5. Native Sequence: Full-length mature human IL-1¦Â with complete biological function.

Storage and Stability
Storage Conditions: Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
Shipping: The product is shipped at ambient temperature. Upon receipt, store immediately at the recommended temperature.
Shelf Life:
- 12 months from date of receipt, -20¡ãC to -70¡ãC as supplied.
- 1 month, 2¡ãC to 8¡ãC under sterile conditions after reconstitution.
- 3 months, -20¡ãC to -70¡ãC under sterile conditions after reconstitution.

Protocol (For Reference Only)
Important: Centrifuge the vial briefly before opening to bring the contents to the bottom. Reconstitute under aseptic conditions.
1. Reconstitution: Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL.
2. Aliquoting: Apportion stock solutions into working aliquots and store at ¡Ü -20¡ãC.
3. Further Dilution: Further dilutions should be made in appropriately buffered solutions.
4. Activity Assay: The biological activity can be measured by cell proliferation assay using murine D10S cells. The ED50 is less than 1.0 pg/mL.
5. Working Concentration: Optimal working concentrations should be determined empirically for each cell type and experimental system.

Precautions
1. Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
2. Reconstitute with carrier protein (0.1% BSA) to maintain protein stability.
3. Stock solutions should be apportioned into working aliquots to avoid repeated freeze-thaw cycles.
4. IL-1¦Â is a potent proinflammatory cytokine; handle with appropriate safety precautions.
5. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)
Q1: How should I reconstitute IL-1¦Â?
A1: Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL. The addition of BSA as a carrier protein is recommended for stability.
Q2: What is the specific activity of this protein?
A2: The ED50 is less than 1.0 pg/mL as determined by murine D10S cell proliferation assay, corresponding to a specific activity of >1.0 ¡Á 10⁹ IU/mg.
Q3: How does this IL-1¦Â compare across species?
A3: Mature human IL-1¦Â shares 96% amino acid sequence identity with rhesus and 67%-78% with canine, mouse and rat IL-1¦Â.
Q4: What is the molecular structure of this protein?
A4: The protein is a single non-glycosylated polypeptide chain containing 153 amino acids with an apparent molecular mass of approximately 17.3 kDa.
