Derivation
The SJSA-1 cell line (formerly OsA-CL) was established in 1982 from the primary tumor of a patient diagnosed with primitive multipotential sarcoma of the femur.

There is amplification of the gene which encodes the p53 associated protein, MDM2.

SJSA-1 cells also exhibit a 15 fold amplification of the gli proto-oncogene.


Background
Complete Growth Medium The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%. 
Subculturing Remove spent medium, add fresh 0.25% trypsin, 0.53 mM EDTA solution, rinse and remove trypsin. Let the culture sit at room temperature (or at 37¡ãC) for 2 to 5 minutes. Add fresh medium, aspirate and dispense into new flasks.
Subcultivation Ratio: A subcultivation ratio of 1:5 to 1:10 is recommended
Medium Renewal: Two to three times per week

Freeze medium: Complete growth medium, 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor temperature
