Nile Red,7385-67-3, IC-03356831

Product Introduction

Nile Red is a fluorescent lipophilic dye widely used for the detection and quantification of intracellular neutral lipids and lipid droplets. It exhibits environment-dependent fluorescence: in non-polar lipid-rich environments, Nile Red emits strong yellow-gold fluorescence (Ex/Em ~552/636 nm), whereas in polar environments it emits weak red fluorescence. This property makes Nile Red a valuable tool for studying lipid metabolism, adipocyte differentiation, steatosis, and lipid droplet dynamics in cells and tissues. Nile Red is compatible with fluorescence microscopy, flow cytometry, and microplate reader-based assays, and is suitable for both live-cell and fixed-cell imaging applications.

Product Features

1. Lipophilic Fluorescence: Strong yellow-gold fluorescence in lipid-rich environments, enabling sensitive detection of neutral lipids.

2. Environment-Sensitive: Fluorescence emission varies with polarity, allowing discrimination of lipid droplets from other cellular structures.

3. Broad Application: Suitable for fluorescence microscopy, flow cytometry, and microplate-based assays.

4. Live-Cell Compatible: Can be used for live-cell imaging of lipid droplets.

5. High Purity: Supplied at 98% purity for reproducible staining.

Protocol (For Reference Only)

Important: Protect from light throughout the procedure. Prepare working solution fresh before use.

1. Preparation of Working Solution: Dilute the stock solution with pre-warmed serum-free cell culture medium or PBS to a final concentration of 200-1000 nM. Note: Adjust the working concentration according to experimental requirements; prepare fresh before use.

2. Cell Staining:

2.1 Suspension cells: Collect cells by centrifugation, add PBS, and wash twice for 5 minutes each. Adherent cells: Discard culture medium, add trypsin to digest cells, centrifuge and discard supernatant, then add PBS and wash twice for 5 minutes each.

2.2 Add 1 mL of Nile Red working solution and incubate at room temperature for 5-10 minutes.

2.3 Centrifuge at 400 x g for 3-4 minutes at 4¡ãC and discard supernatant.

2.4 Add PBS to wash cells twice for 5 minutes each.

2.5 Resuspend cells in 1 mL of serum-free medium or PBS and observe using a fluorescence microscope.

Precautions

1. Protect from light throughout the procedure.

2. Prepare working solution fresh before use.

3. Adjust working concentration according to cell type and experimental requirements.

4. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)
