Recombinant Human CCL25 / TECK, CYT-O15444

Product Introduction

CCL25 is a member of the CC family of chemokines. It is also known as Thymus-expressed (TECK) chemokine because its expression is restricted primarily to the thymus and intestine. In particular, dendritic cells derived from the thymus but not from bone marrow have been identified as a source of CCL25. By binding to CCR9, CCL25 exerts chemotactic effects on thymocytes, macrophages, and dendritic cells. Additionally, CCL25 participates in regulating the development of T cells.

Product Features

1. High Purity: Greater than 97% as determined by SDS-PAGE and HPLC analyses.

2. High Biological Activity: Fully biologically active when compared to standard, with activity determined by chemotaxis bioassay using human monocytes in a concentration range of 1.0-10 ng/mL.

3. E. coli Expression: Produced in Escherichia coli as a single, non-glycosylated polypeptide chain containing 128 amino acids.

4. Low Endotoxin: Less than 1 EU/µg as determined by LAL method.

5. Lyophilized Powder: Supplied as a sterile filtered white lyophilized powder for convenient storage and handling.

Storage and Stability

Storage Conditions: Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

Shelf Life:

12 months from date of receipt, -20 to -70¡ãC as supplied.

1 month, 2 to 8¡ãC under sterile conditions after reconstitution.

3 months, -20 to -70¡ãC under sterile conditions after reconstitution.

Protocol (For Reference Only)

Important: Centrifuge the vial briefly before opening to bring the contents to the bottom. Reconstitute under aseptic conditions.

1. Reconstitution: Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL.

2. Aliquoting: Apportion stock solutions into working aliquots and store at ¡Ü -20¡ãC. Avoid repeated freeze-thaw cycles.

3. Further Dilution: Further dilutions should be made in appropriate buffered solutions.

4. Chemotaxis Assay: Biological activity can be determined by chemotaxis bioassay using human monocytes in a concentration range of 1.0-10 ng/mL.

5. Working Concentration: Optimal working concentrations should be determined empirically for each cell type and experimental system.

Precautions

1. Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

2. Reconstitute with carrier protein (0.1% BSA) to maintain protein stability.

3. Aliquot stock solutions into single-use volumes to avoid repeated freeze-thaw cycles.

4. This material is offered for research, laboratory, or further evaluation purposes. Not for human use.

5. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: What is the biological function of CCL25?

A1: CCL25 binds to CCR9 and exerts chemotactic effects on thymocytes, macrophages, and dendritic cells. It also participates in regulating T cell development.

Q2: How should I reconstitute this protein?

A2: Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL.

Q3: What is the biological activity of this protein?

A3: The biological activity is determined by chemotaxis bioassay using human monocytes in a concentration range of 1.0-10 ng/mL.

Q4: How should I store this product?

A4: Store at -20 to -70¡ãC as supplied for up to 12 months from date of receipt. After reconstitution, store at 2-8¡ãC for up to 1 month or at -20 to -70¡ãC for up to 3 months under sterile conditions.
