DMEM/F12 Medium (without Phenol Red), IC-2209-S
Product Introduction
DMEM/F12 medium is formulated based on DMEM with the addition of the more enriched nutritional components of F12 medium, including a variety of trace elements. It is widely used for the culture of various mammalian cells. DMEM/F12 is also commonly used as a basal medium for the development of serum-free media, and is suitable for mammalian cell culture under low serum conditions as well as clonal density culture.
Phenol red is used as a pH indicator in culture medium to continuously monitor the pH. At low pH, phenol red turns the medium yellow; at high pH, it turns purple; at pH 7.2-7.4, it appears red, which is optimal for cell culture. However, phenol red has certain limitations. Studies have shown that phenol red can mimic the effects of steroid hormones (particularly estrogen); therefore, phenol red-free medium is recommended when working with estrogen-sensitive cells such as mammary tissue. Phenol red can also interfere with flow cytometry detection. Additionally, the presence of phenol red in some serum-free medium formulations may interfere with sodium-potassium balance.
This product contains amino acids, vitamins, inorganic salts, and other components necessary for various cell culture types, but does not contain proteins, lipids, or growth factors. Therefore, this product must be supplemented with serum or serum-free additives before use.
Product Features
1. Enriched Nutrient Formulation: Combines DMEM with the additional nutrients and trace elements of F12 medium.
2. Phenol Red-Free: Eliminates potential estrogenic effects and interference with flow cytometry analysis.
3. Serum-Free Medium Base: Commonly used as a basal medium for serum-free medium development.
4. Stringent Quality Control: Tested negative for bacteria, fungi, and mycoplasma; endotoxin levels <3 EU/mL.
5. Sterile-Filtered: 0.1 µm filtered to ensure sterility.

Protocol (For Reference Only)
Important: Use aseptic technique when handling medium. Warm to 37¡ãC before use.
1. Preparation of Complete Medium: Supplement DMEM/F12 Medium (without Phenol Red) with fetal bovine serum to a final concentration of 5-10% and add antibiotics as required. Mix gently.
2. Medium Pre-warming: Warm the required volume to 37¡ãC for 15-30 minutes before use.
3. Cell Feeding: Aspirate spent medium, replace with fresh pre-warmed complete medium, and return cells to 37¡ãC, 5% CO₂ incubator.
4. Subculturing: When cells reach 80-90% confluency, detach with trypsin-EDTA, neutralize with complete medium, and passage as appropriate.
5. Monitoring: Without phenol red, pH changes are not visually indicated. Monitor cell morphology and growth regularly and replace medium every 2-3 days.
Precautions
1. For research use only. Not for diagnostic or therapeutic use.
2. Handle aseptically to avoid contamination.
3. Do not freeze-thaw. Store at 2-8¡ãC protected from light.
4. This formulation does not contain phenol red; pH changes are not visually indicated. Establish a regular medium change schedule.
5. Supplemented complete medium should be used within 2-4 weeks.
FAQ (Simplified)
Q1: Why choose phenol red-free DMEM/F12?
A1: Phenol red-free medium is recommended for estrogen-sensitive cell types, flow cytometry applications, and studies where phenol red may interfere with detection or cellular responses.
Q2: What is DMEM/F12 commonly used for?
A2: DMEM/F12 is widely used for mammalian cell culture, as a base for serum-free medium development, and for low-serum and clonal density culture.
Q3: Does this medium contain L-glutamine?
A3: Yes. This formulation contains 2.5 mM L-glutamine.
Q4: How should I monitor pH without phenol red?
A4: Without phenol red, pH cannot be visually monitored. Rely on regular medium changes and monitor cell growth and morphology as indicators of culture health.
