DiI-Labeled High Density Lipoprotein, IC-1376

Product Introduction

This product is high density lipoprotein (HDL) labeled with DiI (1,1''''-dioctadecyl-3,3,3'''',3''''-tetraMethyli

-ndocarbocyanine perchlorate), a lipophilic fluorescent carbocyanine dye. HDL is a lipoprotein particle that mediates reverse cholesterol transport, removing excess cholesterol from peripheral tissues and transporting it to the liver for excretion. HDL interacts with scavenger receptor class B type I (SR-BI) and other receptors on the cell surface. DiI-HDL is widely used as a fluorescent probe for studying HDL metabolism, cellular cholesterol efflux and uptake, SR-BI receptor function, and lipid transport mechanisms. Upon binding to cell surface receptors, DiI-HDL can be internalized or undergo selective lipid uptake, with the DiI fluorescence visualized by fluorescence microscopy or quantified by flow cytometry. The DiI fluorophore exhibits orange-red fluorescence with excitation/emission maxima of approximately 549/565 nm.

Product Features

1. Fluorescent HDL Probe: DiI labeling enables visualization and quantification of HDL binding, uptake, and metabolism.

2. Receptor-Mediated Interaction: Preserves native HDL structure for binding to SR-BI and other HDL receptors.

3. Broad Research Utility: Applied in reverse cholesterol transport studies, lipoprotein metabolism, atherosclerosis research, and HDL receptor functional assays.

4. Compatible with Multiple Platforms: Suitable for fluorescence microscopy, flow cytometry, and cell-based assays.

Storage and Stability

Storage Conditions: Store at 4¡ãC, protected from light. Do not freeze.

Shelf Life: The product is stable for 3 months from the date of manufacture when stored as directed.

Protocol (For Reference Only)

Important: Protect from light. Avoid repeated freeze-thaw cycles. Use aseptic technique for cell culture applications.

1. Working Solution Preparation: Dilute DiI-HDL in serum-free culture medium or PBS to the desired working concentration (typically 5-20 µg/mL).

2. Cell Treatment: Add DiI-HDL working solution to cultured cells. Incubate at 37¡ãC for 2-4 hours for uptake studies. For binding studies, incubate at 4¡ãC for 1-2 hours to prevent internalization.

3. Washing: Remove the DiI-HDL-containing medium. Wash cells 2-3 times with PBS to remove unbound DiI-HDL.

4. Detection: Visualize by fluorescence microscopy using a rhodamine/TRITC filter set, or quantify by flow cytometry in the PE channel.

5. Competition Studies: To confirm receptor specificity, pre-incubate cells with excess unlabeled HDL (e.g., 10-50¡Á excess) before adding DiI-HDL.

Precautions

1. Protect from light during handling and storage.

2. Do not freeze; freezing may cause lipoprotein aggregation and loss of receptor binding activity.

3. Optimize working concentration and incubation time for each cell type.

4. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: How should I store DiI-HDL?

A1: Store at 4¡ãC protected from light. Do not freeze.

Q2: What is the typical working concentration?

A2: Typical working concentrations range from 5-20 µg/mL. Optimize for each cell type.

Q3: How can I confirm HDL receptor-mediated binding?

A3: Perform competition studies by adding excess unlabeled HDL (10-50¡Á) to block receptor-mediated binding and uptake. Alternatively, use receptor-deficient cells as negative controls.
