Protocol (For Reference Only)

Important: Handle membranes with clean forceps. Ensure the membrane remains moist throughout the staining and destaining procedure. Perform staining in a clean container.

1. Post-Transfer Staining: After protein transfer, rinse the membrane briefly in deionized water to remove residual transfer buffer.

2. Staining: Immerse the membrane in sufficient Ponceau S Staining Solution to completely cover the membrane surface. Incubate at room temperature for 1-5 minutes with gentle agitation until protein bands become clearly visible.

3. Destaining and Documentation: Pour off the staining solution (may be reused). Rinse the membrane with deionized water several times to remove excess dye and enhance band contrast. Photograph or scan the membrane for documentation.

4. Complete Destaining: Wash the membrane with TBST or PBS for 5-10 minutes with agitation to completely remove the dye. Alternatively, continue with blocking directly as residual dye will be removed during subsequent washing steps. Proceed with standard blocking and immunodetection protocols.

Precautions

1. The staining solution may be reused several times. Discard when staining intensity decreases or solution becomes contaminated.

2. Ponceau S staining is less sensitive than permanent staining methods such as Coomassie Blue; faint bands may not be visible.

3. For PVDF membranes, ensure the membrane is pre-wetted with methanol before staining.

4. For research use only. Not for use in diagnostic or therapeutic procedures.
