Protocol (For Reference Only)

Important: Pre-chill buffer to 4¡ãC before use for optimal cell viability. Protect fluorophore-conjugated antibodies from light throughout the staining procedure.

1. Cell Preparation: Harvest cells and wash once with Flow Cytometry Staining Buffer. Centrifuge at 300-400 x g for 5 minutes at 4¡ãC, discard supernatant.

2. Blocking (Optional): Resuspend cells in Flow Cytometry Staining Buffer containing appropriate blocking reagent (e.g., Fc receptor blocker, normal serum) and incubate on ice for 10-15 minutes.

3. Antibody Staining: Add fluorophore-conjugated antibodies at recommended dilutions in Flow Cytometry Staining Buffer. Incubate on ice for 20-30 minutes protected from light.

4. Washing: Add 1-2 mL of Flow Cytometry Staining Buffer, centrifuge at 300-400 x g for 5 minutes at 4¡ãC, discard supernatant. Repeat wash once or twice.

5. Resuspension and Analysis: Resuspend cells in appropriate volume of Flow Cytometry Staining Buffer. Filter through a 40-70 µm cell strainer if needed. Analyze by flow cytometry.
