1. Cell Preparation: Harvest cells and wash once with Flow Cytometry Staining Buffer. Centrifuge at 300-400 x g for 5 minutes at 4°C, discard supernatant.
2. Blocking (Optional): Resuspend cells in Flow Cytometry Staining Buffer containing appropriate blocking reagent (e.g., Fc receptor blocker, normal serum) and incubate on ice for 10-15 minutes.
3. Antibody Staining: Add fluorophore-conjugated antibodies at recommended dilutions in Flow Cytometry Staining Buffer. Incubate on ice for 20-30 minutes protected from light.
