Dulbecco''''s Modified Eagle Medium (DMEM) was developed based on MEM (Minimum Essential Medium). Compared to MEM, DMEM contains a 2-fold increase in amino acids and a 4-fold increase in vitamins, along with the addition of non-essential amino acids, trace iron ions, and sodium pyruvate. DMEM was initially designed with a glucose concentration of 1000 mg/L (low-glucose type), and a formulation with 4500 mg/L glucose (high-glucose type) was later developed. It is now widely used for the culture of various cell types.
DMEM high-glucose medium is commonly used for fast-growing and loosely adherent cells, hybridoma myeloma cells, cloned cells, DNA-transfected transformed cells, various primary virus host cells, single cell culture, and vaccine production, such as EPO expression using CHO cells and hepatitis B vaccine production.
Phenol red is used as a pH indicator in culture medium to continuously monitor the pH. At low pH, phenol red turns the medium yellow; at high pH, it turns purple; at pH 7.2-7.4, it appears red, which is optimal for cell culture. However, phenol red has certain limitations. Studies have shown that phenol red can mimic the effects of steroid hormones (particularly estrogen); therefore, phenol red-free medium is recommended when working with estrogen-sensitive cells such as mammary tissue. Phenol red can also interfere with flow cytometry detection. Additionally, the presence of phenol red in some serum-free medium formulations may interfere with sodium-potassium balance.

1. Preparation of Complete Medium: Supplement DMEM High Glucose Medium (without Phenol Red) with fetal bovine serum to a final concentration of 5-10% and add antibiotics (e.g., penicillin-streptomycin) as required for specific cell lines. Mix gently by swirling.
2. Medium Pre-warming: Place the required volume of complete medium in a 37¡ãC water bath or incubator for 15-30 minutes before use.
3. Cell Feeding: Aspirate spent medium from the culture vessel, replace with fresh pre-warmed complete medium, and return cells to the incubator maintained at 37¡ãC with 5% CO₂.
4. Subculturing: When cells reach 80-90% confluency, detach adherent cells using trypsin-EDTA, neutralize with complete medium, centrifuge if required, and resuspend in fresh complete medium for seeding into new vessels.
5. Monitoring: Without phenol red, pH changes are not visually indicated. Monitor cell morphology and growth regularly and replace medium every 2-3 days.
