Product Introduction

This product is a high-purity, nuclease-free water treated with diethyl pyrocarbonate (DEPC) and subsequently autoclaved to remove residual DEPC. It is specifically designed for use in RNA and DNA applications where the presence of RNases and DNases must be rigorously excluded to prevent nucleic acid degradation. DEPC treatment inactivates RNase enzymes by covalently modifying histidine residues, providing robust protection against RNase contamination. Following DEPC treatment, the water is autoclaved to hydrolyze residual DEPC into ethanol and CO, ensuring the water is non-toxic and compatible with downstream enzymatic reactions including reverse transcription, PCR, in vitro transcription, and nucleic acid storage. Storage and Stability

Protocol (For Reference Only)

Important: Use aseptic technique and wear gloves when handling to prevent nuclease contamination from skin and laboratory surfaces. Aliquot into smaller volumes for frequent use to minimize contamination risk.

1. RNA Reconstitution and Dilution: Use Nuclease-Free Water to reconstitute RNA pellets following precipitation or column purification, and to dilute RNA samples to desired concentrations for downstream applications.

2. cDNA Synthesis: Use Nuclease-Free Water as the diluent for preparing RNA templates, primer mixes, and master mixes in reverse transcription reactions.

3. PCR and qPCR Setup: Use Nuclease-Free Water as the aqueous component for preparing PCR and qPCR reaction master mixes, adjusting reaction volumes, and diluting primers and probes.

4. In Vitro Transcription: Use Nuclease-Free Water for preparing transcription reaction mixes, diluting RNA polymerases, and resuspending in vitro transcribed RNA products.

5. Enzymatic Reaction Preparation: Use Nuclease-Free Water as the universal diluent for all nuclease-sensitive enzymatic reactions including restriction digests, ligations, and nucleic acid labeling procedures.
