Product Background  

Medium 199 (M199) is a complex synthetic medium originally developed for the nutritional study of chicken embryo cells. It is now widely used for culturing a broad range of mammalian cell types, including primary cells, endothelial cells, and virus production. M199 contains a balanced mixture of amino acids, vitamins, nucleotides, and other growth factors, making it suitable for specialized applications requiring enriched conditions.

- Comprehensive Formulation:  

  - Amino Acids: Includes all 20 essential and non-essential amino acids.  

  - Vitamins: 15+ vitamins (e.g., A, D, E, K, B-complex).  

  - Additional Components: Nucleotides (adenine, guanine, uracil), cholesterol, and Tween 80.  

- Flexible Base:  

  - Available with or without phenol red; HEPES or bicarbonate buffering options.  

  - Glucose-free and low-serum formulations upon request.  

- Physical Properties:  

  - Osmolality: 280¨C320 mOsm/kg.  

  - pH: 7.0¨C7.4 (with 5% COequilibration).  

Usage Instructions  

1. Preparation:  

   - Powdered M199: Dissolve in distilled water, adjust pH to 7.2¨C7.4, and sterilize by filtration (0.22 µm).  

   - Liquid M199: Thaw at 4¡ãC and pre-warm to 37¡ãC before use.  

2. Supplementation:  

   - Serum: Add 5¨C10% FBS or species-specific serum for primary cells.  

   - Growth Factors: EGF, bFGF, or heparin may be required for endothelial cultures.  

   - Antibiotics: Optional 1% penicillin/streptomycin.  

3. Cell Culture:  

   - Use for adherent or suspension cultures at 37¡ãC with 5% CO.  

   - For virus production (e.g., vaccines), supplement as per protocol.  

Quality Control  

- Sterility: Tested per USP/EP standards (no microbial growth after 14 days).  

- Performance: Supports ¡Ý90% viability of human umbilical vein endothelial cells (HUVECs) over 96 hours.  

- Endotoxin: <0.25 EU/mL (LAL test).  

- Consistency: Each lot tested for osmolality, pH, and nutrient stability.  

Precautions  

1. Storage:  

   - Liquid: 2¨C8¡ãC (avoid repeated freeze-thaw cycles).  

   - Powder: Room temperature (15¨C25¡ãC) in a dry environment.  

2. Handling:  

   - Avoid prolonged exposure to light (vitamin-sensitive).  

   - Discard if color changes (pink to yellow indicates pH shift).
